简介
PNKP protein plays a key role in DNA damage repair through NHEJ and BER pathways. Its dual catalytic activity includes removal of the 3'-phosphate group from the DNA backbone and phosphorylation of the 5'-hydroxyl group, ensuring compatibility of DNA ends during extension and ligation. PNKP Protein, Human (His-SUMO) is the recombinant human-derived PNKP protein, expressed by E. coli , with N-His, N-SUMO labeled tag.

Stability Test
The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37℃ for 48h, and no obvious degradation and precipitation were observed. The loss rate isless than 8% within the expiration date under appropriate storage condition.
-
Storage & Shelf Life
Samples are stable for up to twelve months from date of receipt at -20℃ to -80℃. Store it under sterile conditions at -20℃ to -80℃. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.

Shipping
In general, recombinant proteins are supplied as lyophilized powder and shipped at ambient temperature. For bulk packages, the proteins are provided as frozen liquid and shipped with blue ice, unless otherwise requested by the customer.

Protein Description
PNKP (polynucleotide kinase/phosphatase) is a crucial enzyme involved in the repair of DNA and RNA, playing a significant role in maintaining cellular integrity and preventing genomic instability. Its primary functions include phosphorylating the 5' ends of nucleic acids and dephosphorylating the 3' ends, which are essential steps in various nucleic acid processing pathways, including DNA repair, recombination, and RNA processing. Mutations in the PNKP gene have been linked to neurological disorders and syndromes characterized by developmental delays, seizures, and profound sensitivity to DNA damage. Recent research has focused on developing recombinant PNKP proteins to better understand its structural and functional mechanisms, which could provide insights into therapeutic interventions for PNKP-related diseases. Such studies involve the cloning, expression, and purification of PNKP to characterize its enzymatic activities and interaction with other cellular proteins. As a result, understanding PNKP's role in nucleic acid metabolism not only sheds light on fundamental biological processes but also has significant implications for targeted gene therapy and novel treatment strategies for genetic disorders associated with DNA repair deficiencies. The continued investigation into the properties and functions of recombinant PNKP thus represents an important frontier in molecular biology and medicine.
